The Strangetown Hotel is the primary setting for The Sims 2. The player's sim becomes the manager of the hotel and is tasked with maintaining and improving it to achieve a Hotel Score of 100%. It is the largest building in Strangetown as well as the most frequently visited for its many venues. Inside the Hotel are five functioning guest rooms, one of them being a Penthouse which remains occupied by one of three antagonist characters until the player completes the main story. In addition, there's a pool on the Sun Deck, two lounges, a restaurant, a spa and gym, a casino, an art gallery, and many other rooms. All but the Small Hotel Room, Penthouse, and some essential rooms in the Hotel (such as the Manager's Suite and the Reactor Room) must be built by the player.

The main storyline revolves around the hotel. There are, in total, three antagonists who stay in the Penthouse; Frankie Fusilli, Ava Cadavra and Optimum Alfred. Each of them causes some sort of problem that the player must solve in order to progress through the story. Frankie uses the Hotel as a vehicle for money laundering and attempts to assassinate Honest Jackson in an attempt to take political control in Strangetown; Ava uses the player to bolster her cult and resurrect the Prime Heifer; and Optimum Alfred attempts to take control of Strangetown with a robotic, iron fist. The player is assisted in defeating each of these antagonist's by Penelope Redd, Horus Menhoset IX, and Emperor Xizzle, respectively.

There are many locations within the hotel. In addition to entertainment facilities, there are five functioning guest rooms and a handful of basement-level rooms. The Hotel contains many facilities which can't be found elsewhere in Strangetown, making it a popular location for sims to spend time. Entrances to the hotel are located in the Main Lobby and at the back of the Atrium, and it's the only building in Strangetown which has more than one entrance. Listed below are the four "hub" areas of the Strangetown Hotel which provide access to the other areas of the hotel:

Throughout the Hotel, various sims are employed in certain positions. Some of these sims are implied to be of a higher ranking than the position they appear to hold. For example, Futo Maki is heavily implied to be the owner of the Asian Emporium by the introduction text that appears when the room is built. Below is a list of employees in the hotel in alphabetical order. Note that most of them do not have a real name, instead being given a title which reflects their position.
